The Role and Department
The Department of Biosciences is composed of 62 academic staff and research fellows; around 50 research and technical support staff; over 90 Research Postgraduate students and over 500 Single Honours undergraduate students, and also teaches students from the Natural Sciences programme. The Department is committed to an ethos of research-led teaching at both BSc and MBiol levels, and was ranked 5th in the UK for Biological Sciences Departments in the Complete University Guide 2024.
Our basic science underpins applications across each-and-every sector of the bio-economy, including Biotechnology, Ecology and Agritechnology. Research programmes are grouped in the following areas: Molecular Plant Sciences; Biomolecular Interactions; Ecology, Evolution and Environment; and Animal Cells and Systems. These groups organise focused seminar series, Research Away Days, manage joint research facilities, oversee mentoring, internal peer review and serve to nucleate larger research projects. Along with our excellent research facilities such as bioimaging, proteomics, metabolomics and genomics we offer a high quality and supportive environment for our research staff.

The Role
Join us as a Post-Doctoral Research Associate at the newly established Centre for Programmable Biological Matter.
At the Centre for Programmable Biological Matter, we are interested in understanding, designing and building natural and artificial biological nanomachines. We look at the amazing variety of such machines in nature, including in our own cells ask - can we design artificial versions of such machines to carry out processes not available or even not possible in nature? The ability to design functional, dynamic proteins will revolutionise medicine. It will mean new capabilities in vaccines and drug including nanoscale medicine that can be programmed and can move. We have established the Centre of Programmable Biological Matter as to spearhead research in this area.

Project description - 'Developing Artificial Protein Cages for Medicine'.
Inthis project, you join our multidisciplinary team and will lead a project to design and produce an artificial, dynamic protein cage-based vaccine able to stimulate humoral responses, gain cell entry, and open within the cell to release cargoes that further stimulate immune response. This will primarily involve two work-packages, a) Protein-Cage Design and Engineering, and b) Protein Construction, validation and testing. The work is funded by the prestigious Academy of Medical Sciences.
We have already created and characterized a stable artificial protein cage, TRAP-cage, resembling a small virus capsid with controllable dynamic properties. This means the cage can be programmed to open on demand. We also showed that folded proteins can be displayed in multiple copy number, on the exterior of the protein while folded proteins and peptides can be placed into the lumen of the cage - foundational results for vaccines and drug delivery respectively. These cages are highly stable but the conditions under which they open can be programmed, depending on the design. Programmed opening conditions shown to date include in the cytoplasm of the cell and we have shown that cages carrying cargoes can enter cells and open to free cargo. We have also demonstrated cages responsive to low pH. We have identified Lassa Fever as a suitable vaccine target. Its reach is expanding due to climate change and there is currently no human vaccine.
The location for this role will be 100% on-site in our newly refurbished labs with state-of-the art equipment, located in the Department of Chemistry, Durham University.

Experiment and Discover: Apply molecular biology and biochemistry techniques to craft and evaluate advanced systems, pushing the boundaries of what's possible.
Explore and Confirm: Master the art of cryo-electron microscopy to isolate, understand, and validate the structures of the systems you develop.
Collaborate and Advance: Take the most promising dynamic nanostructures to the next level in cellular studies, working alongside experienced collaborators.
Key responsibilities:
This post is fixed term for 24 months with a flexible expected start date prior to April 1st 2025.
The post-holder is employed to work on research/a research project which will be led by another colleague. Whilst this means that the post-holder will not be carrying out independent research in his/her own right, the expectation is that they will contribute to the advancement of the project, through the development of their own research ideas/adaptation and development of research protocols.

A good first degree in Biological sciences, including but not limited to protein chemistry, biochemistry, biophysics and structural biology, computational biology, computer sciences, natural sciences, mathematics. A PhD (or be close to submission) in structural biology, protein engineering, physical virology or a related subject.
Experience
A proven track record in protein biochemistry.
